Dosing Protocol

Kisspeptin-10 is administered via subcutaneous injection once daily throughout the active cycle.

Evening dosing is preferred, Kisspeptin-10 drives pulsatile GnRH and LH release, and aligning the injection with the evening LH surge window produces the strongest downstream testosterone response. The peptide has a short half-life of approximately 30 minutes, so timing relative to the natural LH pulse rhythm is more important than rigid clock timing.

Do not use Kisspeptin-10 if you have hormone-sensitive malignancy, active pituitary disease, or are currently on GnRH agonist or antagonist therapy, these compounds act on the same receptor pathway and will block or compete with Kisspeptin-10's mechanism. Monitor LH, FSH, and total testosterone at baseline and at weeks 4 and 8 to assess HPG axis response.

Kisspeptin-10 acts at KISS1R receptors on hypothalamic GnRH neurons, the top of the HPG axis, to drive pulsatile GnRH release and downstream LH, FSH, and testosterone secretion. Published clinical research has documented significant LH pulse amplitude increases and measurable testosterone elevation following subcutaneous Kisspeptin-10 administration at 100–1,000mcg in both healthy and hypogonadal subjects. The stepwise titration reflects the peptide's receptor sensitivity profile, starting low allows the HPG axis to respond progressively rather than saturating KISS1R receptors at first use. The 4-week rest period allows hypothalamic receptor sensitivity to restore before the next cycle. At 3mL BAC, concentration is 3,333mcg/mL, intro and titration draws are approximate and require a 3mL micro needle syringe.
In biohacking forums, 250–500mcg once daily in the evening is the most commonly reported pattern, with some users reporting measurable improvements in libido and morning erection frequency within the first 2 weeks at the titration dose. Use alongside testosterone support protocols and PCT frameworks is noted across community discussions. These are community-derived patterns, not clinical.

Administration Timing

Kisspeptin-10 is injected subcutaneously once daily in the evening. Timing relative to the natural LH pulse rhythm is the primary administration variable.

  • Evening injection, 8–10 PM preferred: The hypothalamic LH pulse generator is most active in the late evening and early sleep period. Injecting during this window aligns the Kisspeptin-10 peak with the body's natural GnRH pulse rhythm, producing a stronger and more physiologically coherent LH and testosterone response.
  • Consistent daily timing: Kisspeptin-10 drives pulsatile hormone release, irregular timing disrupts the LH pulse pattern it is designed to support. Hold the same injection window within 30 minutes each day throughout the cycle.
  • Bloodwork at baseline, week 4, and week 8: LH, FSH, and total testosterone are the primary markers to track. Baseline values before the first injection establish the reference point; mid-cycle and end-cycle checks confirm HPG axis response and guide dose decisions for subsequent cycles.
  • Switch syringe type at week 4: Once you advance to the 500mcg maintenance dose (~15 units), a standard U-100 insulin syringe is accurate and appropriate. The micro needle is only required for the intro (~3 units) and titration (~7.5 units) phases.
  • Rotate injection sites each session: Abdomen and outer thighs. With once-daily dosing across an 8-week cycle, systematic rotation prevents site irritation.

Notes & Best Practices

  • Kisspeptin-10 works upstream, effects are driven by your HPG axis response: The peptide triggers the hormonal chain; the magnitude of the response depends on the functional capacity of your GnRH neurons, pituitary, and testes. Bloodwork is the only reliable way to confirm the protocol is producing the intended LH and testosterone response.
  • Do not combine with GnRH agonists or antagonists: Compounds like leuprolide, cetrorelix, or degarelix act directly on GnRH receptors and will block or override Kisspeptin-10's mechanism. These combinations are contraindicated.
  • Libido and arousal effects are hormonally mediated: Unlike PT-141, which acts centrally and independently of hormone levels, Kisspeptin-10's effects on desire and arousal are downstream of LH and testosterone elevation. Effects build across the cycle as testosterone rises, not within hours of a single injection.
  • Complete the titration phase before assessing response: The intro and titration phases exist to allow KISS1R receptor sensitivity to calibrate progressively. Jumping to 500mcg at first use risks receptor oversaturation and a blunted subsequent response. Follow the stepwise protocol.
  • Discard at 28–30 days post-reconstitution.

High Dosage Side Effects

Taking too much Kisspeptin-10 can increase side effects. Signs your dose may be too high:

  • Injection site redness or irritation: The most commonly reported above-range sign across Kisspeptin research. Reduce dose and rotate sites more consistently if persistent beyond 24 hours.
  • Nausea or mild gastrointestinal discomfort: Reported at higher doses in clinical trial subjects. Reduce to the previous titration step and hold for one week before advancing again.
  • Headache post-injection: Dose-dependent and typically resolves within 2–3 hours. Reduce to the previous dose step if it occurs consistently across multiple injections.
  • Excessive testosterone elevation symptoms: Acne, oily skin, irritability, or elevated haematocrit on bloodwork. Reduce dose and recheck bloodwork within two weeks before continuing.
  • LH or testosterone overshoot on bloodwork: Values significantly above the upper reference range indicate the dose is producing a larger HPG response than intended. Reduce to 250mcg and recheck at the next scheduled bloodwork point.

If any of these occur, reduce to the previous titration dose and hold for one week before reassessing.

Low Dosage Side Effects

A dose may be too low if expected effects are absent after consistent use. Common indicators:

  • No improvement in libido or energy after 4 weeks at 500mcg: Confirm bloodwork before adjusting. If LH and testosterone have not risen measurably from baseline after 4 weeks at maintenance dose, advancing to 1,000mcg (~30 units) for the remainder of the cycle is within the documented research range.
  • No change in LH or testosterone on mid-cycle bloodwork: The most objective low-dose indicator. Flat LH alongside absent subjective effects after a full titration phase confirms the dose is below the individual's activation threshold, advance to 1,000mcg with bloodwork monitoring.
  • No injection site response at all: Mild transient redness at the injection site is common at therapeutic doses. Complete absence alongside no other effects may indicate underdosing or technique issues, confirm micro needle accuracy for intro draws before advancing.

If these indicators persist after a full 4-week maintenance phase at 500mcg with confirmed correct technique, advancing to 1,000mcg (~30 units) for the second half of the cycle is appropriate with bloodwork monitoring at the end of the active phase.
